LC-MS / MS

The LC-MS/MS analyses of the platform are performed on Bruker’s nanoElute/timsTOF Pro package.

The timsTOF Pro has 2 ion mobility modules placed in series, the first one allowing the trapping of eluted peptides over 100 ms while the second one ensures the ion mobility separation of the previously trapped peptides. The speed of qTOF downstream of the ion mobility is exploited by the PASEF technology which allows an MS/MS analysis frequency of about 100 Hz.

This results in a more exhaustive (duty cycle = 100%) and more resolving analysis of the peptides (additional separation of the peptides according to ion mobility) as well as a higher sensitivity.

The nanoElute is a UHPLC allowing reproducible peptide separation. The coupling with the timsTOF Pro is done through the Captive Spray, a system that confines the spray as close as possible to the input of the mass spectrometer and allows a gain in sensitivity.
